چکیده

Newly-developed gel isoelectric focusing techniques provide for rapid and correct identification of human hemoblobins. With this technique, we examined samples previously identified by the National Hemoglobinopathy Standardization Laboratory of the Center for Disease Control. A wide variety of both normal and abnormal hemoglobins was included in the 134 different specimens examined. The isoelectric points (pl's) were precisely determined; no discrepancies with results by standard techniques of identification were found. The method is accurate and precise. It is proposed as a powerful adjunct to other laboratory methods suitable for use in a hemoglobinopathy reference laboratory or specialized hematology service laboratory.

A Simpler and Faster Version of Two-Dimensional Gel Electrophoresis Using Vertical, Mini SDS-PAGE Apparatus

We have modified one of the most useful methods of protein separation; namely, two dimensional gel electrophoresis (2-DE). This modified version of 2-DE is not only simpler and easier but also faster than all the currently available methods. In this method, isoelectric focusing is carried out in the first dimension using a vertical sodium dodecyl sulfate polyacrylamide gel electrop...

Isoelectric Focusing ofHuman Hemoglobin: Its Application to Screening, to the Characterization of7O Variants, and to the Study of Modified Fractions ofNormal Hemoglobins

Isoelectric focusing on slabs of acrylamide could not be separated from HbS by gel was adapted for the screening of abcellulose acetate electrophoresis. The normal hemoglobins, the characterization characterization technique of electrofocusof 70 human variants, and the study of ing allowed us to distinguish between minor fractions of normal hemoglobin. The most variants. Only one mutant, Hb scr...

Characterization of Nucleoprotein Extracted from Human Influenza A Virus Cultured in Two Different Cell Lines

Background and Aims: Influenza virus nucleoprotein (NP) has the capacity to be used as subunit vaccine, but little is known about the impact of different cultures on its structure. In the present study we aimed to evaluate and compare the Isoelectric focusing (IEF) property of extracted viral nucleoproteins derived from Madin Darby canine kidney (MDCK) cell line and embryonated chicken eggs (EC...

Development of non-gel-based two-dimensional separation of intact proteins by an on-line hyphenation of capillary isoelectric focusing and hollow fiber flow field-flow fractionation.

A rapid, non-gel-based, on-line, two-dimensional separation method is introduced for proteome analysis. Protein fractionation was carried out by first exploiting the differences in their respective isoelectric points (pI) in a Teflon capillary using isoelectric focusing (IEF), followed by a molecular weight (MW)-based separation in a hollow fiber by flow field-flow fractionation (FlFFF). The me...

Identification of Fish Species by Thin-layer Polyacrylamide Gel Isoelectric Focusing

Conventional electrophoretic techniques for the identification offish species are limited in the resolution and reproducibility needed for the reliable identification of fish species. This paper describes the potential of a high resolution protein separation technique, thin-layer polyacrylamide gel isoelectric focusing (IEFl, as a new means of identifying fish species. Sarcoplasmic protein patt...
